updating by painting paneling in den of 60s house

Do I need to prime the paneling before painting or is there a paint that will do the job in one coat?

I have painted paneling. You should prime or will go through more paint than necessary. Also if you want to get rid of ridges, fill them with premixed drywall mud. Once dry, lightly sand so it's smooth. Then prime and paint.
