My vanity is 21”deep 34”wide, will this vessel fit? If so where do you recommend the drain hole & faucet hole to be located?

We recommend you to do the drain and faucet holes when the vessel sink arrives. Also the standard height for a vessel vanity goes from 30 inches up to 35 inches.
