What are your DIY tests/experiments for pathogens?

Most of us can look for bacterial ooze and perform bacterial streaming tests. Most of us can look for symptoms to help narrow down what is wrong with our plants ie, cankers, spots, yellowing.... or at least to narrow it down to biotic/abiotic and virus/bacterium/fungus.

I would like to try to experiment with simple tools like a common microscope, dissecting scope, etc to find out if there are any other ways to diagnose disease at home.

It costs at least $80 per sample to get pathogens Dx'd at state agriculture facilities or land-grant universities with Ag. dept.'s.
