I would do a conventional 3-in-a-row layout: vanity, toilet, and shower. That is, a standalone shower to replace the tub, vanity in same location, but turn the toilet 90 degrees such that it is parallel to the shower. I would put the makeup area in the space currently occupied by the shower and would have that countertop at desk-height.

WHY do people think a moist room is where one should sit and do makeup and hair?? That is counter indicated. Removing the pocket door wall will give you space. Walls take up footage for sure but that is where your make up should be applied so I would rethink removing that wall. If you abandon the tub and put in a shower that you can stand in w/o touching the walls when you turn around you might find room for a sink and vanity and still have the alcove for make up. Yes all vanities ought to be desk height or kitchen counter height depending if you are standing (sink) or sitting (make up). It would be unusual for a person to be actually getting much use out of those jacuzzi tubs everyone was putting in two decades ago. They are a huge space sucker. Assuming your home has a tub elsewhere do you really think it's worth the sq. footage??

I think coastal_vibe has the perfect idea for layout. Counter height for vanity and desk height for makeup. You can turn the makeup area into a special place with a backlit led mirror and maybe some pretty wallpaper. Allow 42-48" width for a comfortable size shower and 36" for opitmal space around the toilet for cleaning. When I did my bathroom vanity, I actually had them flip the sink bases upside down so I had 2 30" drawers at the bottom for incidentals and plenty of room inside for organization.
