anyone trying micro budding?

I'd like to know if micro budding (esp. citrus) is something a person could do at home. Has anyone tried this, or any similar techniques?

    Never tried myself (had to look up what micro budding is) but appears to work with citrus: https://ucanr.edu/repositoryfiles/ca3611pCR-16-70788.pdf

    I am trying to grow individual buds of avocado, fig, and a few other trees in my garden in vitro and so far it looks promising.

    Could you explain what you mean by "grow individual buds"?

    Basically cut the bud and grow it in culture media. It's a method of micropropagation (more specifically culturing the apical meristem) but I wanted to see if it can be done at home. So far it's looking good!

    I'm very curious to know some details about the way you are doing this. For example, are you using a binocular microscope when you cut out the apical meristem, or do you have a cruder way of doing this. And how are you sterilizing the stem w/buds before you cut them out. What kind of culture are you using and in what kind of container...these are the types of things I'd like to know more about.

    Are you using grow lights and a heating mat also...those I have already.

    If you're interested, I just found an 1992 paper with some details, including how to make different kinds of culturing media. It's a little more than I want to get into, ie: the stock salt solution consists of 11 different chemicals, but the detail is there. I'm guessing you are purchasing culturing medium from somewhere.

    This paper also mentions the sterilization of the stem sections:

    30 secs in 70% alcohol, then 15 mins in 2.5% calcium hypochlorite. Then wash 4 times with sterile distilled water to remove excess hypochlorite. Are you doing something similar?

    If you're doing this without having to use a microscope that would make it much more interesting to me. I originally read the ucanr.edu paper you linked to, but the requirement of aseptic conditions and a binocular microscope kept me from following up on it.

    I'll try to get some pictures tonight. I had been experimenting with micropropagation and my understanding was researchers are isolating and culturing the meristem in labs while hobbyists typically culture the section of the stem with the node, so why not try "fusing" the two techniques?

    I do not have a microscope or laminar hood so it's definitely crude. I found it nearly impossible to sterilize a full bud (I use NaDCC instead of alcohol or hypochlorite since it seems less harmful to the tissue) so what worked for me was using sharp disposable scalpels, cut away as much of the bud and primordial leaves with magnifiers, down to about where the meristem lives. Sterilize that in NaDCC for a few hours then plate.

    Fig was great to start with since the buds are large and forgiving. The only advantage to this technique would be the possibility of getting to the actual meristem with a microscope to produce virus-free clones. Otherwise, I imagine you'd get the same result with a node section of the stem and rigorous disinfection. I like doing it my way just for the fun of it : ).
