Option 2 looks like the better option in terms of layout, mostly because by moving the Laundry room to align with the bathroom, so all the plumbing is concentrated in one area. Also having laundry room separated from the Master Bedroom wall will be better as you don't want to hear the sound of the machines if there is any late-night laundry sessions.

Regarding the layout for the Master Bedroom, I would invert the bed so it's actually on the wall facing the hallway. That creates a better walkway and more space in the room. You can add a seating area to the front of the bed, and keep the dresser. Where the window is you can add a little chaise or an additional desk if there are two adults who work from home.

I vote for layout 2 also for the same reasons Magy Interiors LLC mentioned. I would make small modifications to the master bathroom:

- move the toilet in a more private location

- extend the vanity

- the blue rectangle is a 'wasted space but maybe you could make it a small linen closet for towels, etc

EDIT: You have plenty of room to move the toilet to the North and still very private. That would allow you to make the linen closet wider.

Do you mind drawing the contour of the current layout so we know exactly what is an extension and what is not?

Yes! Great idea, will upload when I get home but basically everything upwards of the master bedroom is an addition (so master bath, patio and office/bedroom).

Is the back of the house just a straight line .i.e. everything above the blue line is the extension?

One other change you might consider is to add a cabinet inside the guest bathroom using space from the bedroom 4 closet. If you find it becomes too small then I would let go of the closet to the right (accessed from the hall way).

EDIT: I am not sure if you have room for it but if you do, I would add a little counter top beside the sink. I can become handy when you hold something wet in your hands.

I modified the master closet and the master-ensuite. I added a cabinet inside the bathroom to the right of the door. I put a nice dresser (with maybe an art above it) in your closet. It is up to you but I believe you don't need a door to your closet.

The other changes have already been mentioned in my previous comments.
