Above basement room, below kitchen floor??

I have a crawl space under most of my home but there is also a full height room and the garage is next to it. The house is built on a small sloped lot. The house is single story exceot for thethis room and garage which both have living space above. The side walls are cinder block. Between crawl space on one side and garage in the other. The front wall is the brick exterior if the home. The back wall which is in the utility room behind this room, separated by a hard cement board wall and door. That room is semi below grade and also cinder block. The room is approx 12x16. There is a large crank out style window in the front wall. Currently there is 8x8 Armstrong tiles as ceiling. Basically pressed paper. Some are stained from previous leaks, some are screwed in from repairs, etc etc. i want to tear them down and replace with regular sheet rock. Do i need insulation between that ceiling and the kitchen above. The room is heated with a single 2x12 register and a cold air return. It has a concrete floor. The room is always a several degrees cooler than upstairs. Any suggestions would be appreciated.
