I have a couple of questions that may help get this started. You mentioned removing Jacuzzi, but are you still going to want a tub? Or getting rid of the tub alltogether and just keeping shower? That seems like a smallish shower for a large ensuite, so even if you lose jacuzzi/tub, are you wanting to open the shower up more?

I would first suggest working with a professional who can better evaluate your home in person for these moves and changes so you can keep costs lower and keep structure in mind. Taking out exterior windows/moving plumbing will probably start to incur much higher costs than just a remodel/upgrade of some basic areas/paint/finishes/closing off space.

For the closets, you mentioned moving them. You don't want to lose good closet/storage space, and this space is also between the bedroom and bathroom so not an area that would be good to repurpose for the office. We do have some very experienced home remodelers on here but I think some more info on how you see things functioning would help them give suggestions. It could also help to add some pictures of the rooms are they currently sit here in the comments.

I would go with a shower only. Put it on the window wall and put a double vanity opposite the shower. If the window is too low for privacy purposes, update it with a clerestory window that goes the full length of the shower. Will bring in much needed light to the bathroom.

If getting rid of the angled walls are in the budget I would advise it.
