Noise mitigation between floors

Hello...One bedroom on main floor sits directly over another bedroom in a finished basement. The floor (100+ years old) between the two rooms does not muffle noise well. I am thinking of laying wood flooring on top of the existing wood flooring to decrease the noise transmission between floors. What are thoughts about the likelihood of wood flooring on top of existing wood floor decreasing the noise and squeak factor?

Also, the HVAC has been updated so there is an unused vent that I suspect carries some noise (well enough to hear conversations in room below). If it is unused can I simply stuff it with insulation without causing issues? There are still the many, many squeaks though.

Open the ceiling and fill it with mineral wool.

Noise, minimal at best. Squeak factor, very well may increase it considerably.

The only way to achieve your goal by adding a floor would be to isolate it from the substrate. A floating floor laid over sound control matting could decrease sound and impact transmission. If the existing floor has squeaks, those should be eliminated before adding a new floor over top. G&S has the right idea. After removing and insulating the ceiling below, look into sound-dampening drywall, ideally as part of a sound control system. While the drywall ceiling is out, you may be able to determine where squeaks are coming from and add some fasteners from underneath.
