I think the patio doors give a lot of light to the space . Could you live without a window in your office? The mud room/ laundry can go in that area at the top of the plan . Is there plumbing in there? The HVAC really needs to stay where it is since moving that is very expensive. I would probably put your office where you have that wall across from your storage racks I think. Then instead of bunks I would furnish the rest of the space with sofa beds and a TV where the kids could go when you are using the FR for your TV. The fact that there are no other windows is the reason for keeping the patio door and if the sleeping area is left open then that is the egress from the sleeping space .I would much prefer the laundry on the main floor but that is something you need to decide.

I think the laundry room on main floor is ideal. But do you have room for a 2nd set in the basement? I live on a farm. It doesn’t matter where that washer is, the entry ways get dirty, LoL. I have 3 stacked sets in the basement and one stacked set on my main floor. They are all Mieles that, with the exception of the main floor set, are old sets I got off Craig’s lists, eBay, etc. there are a few machine freaks on this forum ;)

If you do put a set in the basement you could get a condensing type dryer and not need to worry about ducting. I’ve never use one but many are happy with theirs.
