Are you talking about shortening a window or the shower stall? What wall behind the toilet would you want to remove? Why bother to put the W/D in a closet? They don't look that bad and you may not have to shorten the shower stall if you can just stand them at that end. With no closet doors to open, you can adjust the vanity a bit to the right. If I were going to hide anything in a closet it would more likely be the toilet.

I was talking about shortening the window, not shower.

That short black wall near the doorway is next to the toilet. I wasn't sure how to make the layout in the software program with a wall that extends past the doorway.

I don't want to walk into the bath and see a stackable washer/dryer on display. I was just going to put a door in front of it.

That wall is 13' long so I could easily have a 48"-60" vanity and have space on that wall to open the doors to the washer/dryer. And the shower will still be at least 4x5 which I thought was plenty big for a guest shower.

This is our only bath downstairs so will be used throughout the day by us and our kids as well as guests. There are 3 baths upstairs as well for each bedroom (for perspective).

Is the guest bedroom downstairs? If all bedrooms and other baths are upstairs, you might consider eliminating the shower here and dividing the space into a nice powder room and laundry room, separate from each other.

The last place I would put a laundry is in my guest bathroom. Maybe explain a bit about the reasoning and BTW laundry is much more convienient on the main floor , it requires one trip down and one trip up instead of up and down for every load.

So bottom line, we are putting a stackable washer/dryer in this bathroom.

The only thing I guess I really need advice on is the positioning of the vanity and toilet. Does this layout look good? Should i move the toilet? How should the vanity be positioned? Up against the wall or centered in front of the toilet?
