The ensuite thing doesn’t sound like a big deal at all. Unless a plumbed fixture must be moved or your walls are 18” thick brick or concrete.

The half bath - not nearly enough info provided. Is there already a small room/closet that will be used? Or do you have to create a new space from another space? Is there plumbing nearby (both incoming and outgoing plumbing) already?

Are there other bedroms that use that bath? A hall bath is much more versatile than an en suite. It can serve as the public powder room as well, without the 40K+ addition of creating a new powder room.

littlebug zone 5 Missouri, thanks for your comment! There is actually a laundry room next to the bathroom so it goes like this down the hallway -- laundry room with sink, and to the right of that, a bathroom with shower and then to the right of that, the guestroom. Turns out, I think there's some space in the walls between the laundry room and bathroom so I think I can put the powder room in there! So the 1/2 bath (powder room) will be on the other side of the bathroom - does that make sense. I'm okay with losing space in the laundry room such as if I have to lose the sink in there or have to do stacked laundry. The question is how to configure it.... I have wanted to an ensuite like this since I got the house in 2006 and I am now doing a full remodel.

I would not sacrifice space in my laundry room to the point of having to stack laundry machines and give up the sink just to make a cushier nest for guests.
