Random thoughts:

- Overall thought 1: Open the living - primary -closet into one large space ... use this for dining and living. Use the current dining space as a third bedroom, which gives all three bedrooms good access to the bathroom.

- Overall thought 2: Move the kitchen to the back of the house and use the galley kitchen space for your dining table. It'd make for a skinny dining room, so consider banquette seating.

- This is a one-story house, right? So nothing under the stairs? For sure use that under-stair space for something. Widen the bathroom a bit? If you move the dining room to the kitchen space, it could be shelves for dishes, etc.

- Think twice about a French door. I hate mine because of the inswing space -- wish I had a slider /glider. They make them with mullions that look like French doors.

It is one story so stairs go to basement.

If you don't care about losing a bedroom I would make the dining room the primary bedroom and make the back bedroom next to it the walk in closet. I would make the current kitchen a ensuite bathroom, take down the current wall for the current primary bedroom and closet and make a kitchen there open to living room with dining between. The current hightlighted closet areas i would make a hall into the back bedroom areas. then build into the back bedroom for the hightlghted bedrooms closet. OR if you don't want an ensuit bathroom flip the living room to the right side put the kitchen on the left and make the existing kitchen a eat in or small dining area.

Maybe what you said Design Interior South but make highlighted area the hall/open office area with built-ins/storage and make existing kitchen the primary closet?

It is a small room and if youre good with only having one bedroom then you can use the hightllighted room as an office. If you need spill over guest sleeping have a murphy bed installed in there for when you have a guest. IDK the age of this home but my guess is older and what you are seeing possibly not the original floorplan. I doubt the current primary and closet was that. Anyway I did a quick sketch the same scale you shared and drew out one idea. I eliminated the door in the now primary bedroom to the outside because who wants people tromping through their bedroom and located an exterior side door in the living room instead.

Variation of Design Interior South's suggestion:

Keeps three bedrooms (one shown as an office), adds a bath for the primary bedroom (don't know if plumbing can work there, though). If the ceiling is joists, not trusses, and they run horizontally on this plan, opening the living/dining/kitchen area will require beam(s). I also switched a window with the door on the front for easier furniture arrangement and added a fireplace and flanking windows to the left living room wall. Possible??

Or one more option.....keeping your kitchen on the plumbing side (old kitchen) keeping only one bath but continuing a hall to the back door. IDK if there is a yard back there ??? Seems like there must be since the dining room had an exterior door, If not then I wouldn't worry about a second exit but if there is I assume you will want to have access to it other than exiting through the front door and walking around the house .
